The in vivo micronucleus assay in mammalian bone marrow and peripheral blood. A report of the U.S. Environmental Protection Agency Gene-Tox Program

Abstract

The protocol recommended for the micronucleus assay in mammalian bone marrow has been revised and simplified. The number of sample times has been reduced to one or two, depending upon the dosing protocol. The minimum number of cells to be scored per treatment group has been increased to 20 000 to increase the ability of the assay to detect a doubling of the control micronucleus frequency. Use of both male and female animals is recommended. Scoring of micronuclei in polychromatic erythrocytes of peripheral blood is included as a variation of the bone marrow assay. Published data on chemicals tested by the micronucleus assay have been reviewed and are summarized.

      The DF (%) was calculated based on the number of cells with a tail versus those without a tail. The MN test was performed according to the US Environmental Protection Agency Gene-Tox Program guidelines (Mavournin et al., 1990). The bone marrow from both femurs was suspended in fetal calf serum and the smears were prepared on clean glass slides.

      OxBC was evaluated for in vivo clastogenic activity and/or disruption of the mitotic apparatus by detecting micronuclei in polychromatic erythrocytes in CD-1 mouse bone marrow. Experimental procedures complied with OECD, ICH and EC Guidelines, US EPA Pesticide Assessment Guidelines, and the Japanese Guidelines on Genotoxicity Testing and recommendations published by the US EPA Gene-Tox Program and the Japanese Collaborative Study Group for Micronucleus Testing (Environmental Mutagen Society, 1990; Mavournin et al., 1990). The vehicle control was a solution of N-methyl-2-pyrrolidone, polyethylene glycol 400 and propylene glycol prepared in the ratio of 1:2:2, respectively.
