      Without metabolic activation, strain TA100 is non-responsive and strain TA1537 is only responsive in about a quarter of all cases. This is consistent with published results: most mutagens in TPM of MS have been reported to require S9 metabolic activation, and Ames endpoints without S9, even when responsive, have been reported to yield significantly lower responses than corresponding endpoints with S9 (DeMarini, 1983; DeMarini et al., 2008; Rickert et al., 2007). In vitro endpoints with response rates at or below the few-percent-level will not be considered further, thus the Ames TA100, TA102, TA1535, TA102 +S9 and TA1535 +S9 endpoints are removed from the analysis due to absence of data to facilitate further statistical treatment (Fig. 1).

      The plate incorporation version of the Ames assay was used and applied to TPM only, because this assay version is not capable of detecting the mutagenic potential of GVP. The Ames assay can discriminate the mutagenic activity of TPM from different types of cigarettes (Roemer et al., 1998) with different filter ventilation, filter efficiency, paper porosity (Tewes et al., 1999), and tobacco types as well as cigarette smoke fractions (DeMarini, 1983). In order to mimic mammalian metabolism, the TPM-DMSO suspensions were also tested in the presence of a metabolic activation system.

      Cigarette smoking is the single most preventable cause of debilitating illness and premature death (US, 1989). Cigarette smoke constitutes 4500 chemical compounds and some of them are known to be mutagens (DeMarini, 1983; Obe et al., 1984). Investigations reported that there are 1.1 billion smokers worldwide and 182 million (16.6%) of them live in India (WHO, 1997; Gajalakshmi et al., 2000; Jha et al., 2002; Shimkhada and Peabody, 2003).
