Inhibition of phenylalanine ammonia-lyase by cinnamic acid derivatives and related compounds

Abstract

Thirty-five derivatives of cinnamic acid and related compounds were tested for inhibition against phenylalanine ammonia-lyase (PAL) derived from sweet potato, pea and yeast. Caffeic and gallic acids showed inhibition against PAL originating from higher plants, but not against yeast PAL. In contrast, yeast PAL was specifically inhibited by p-hydroxycinnamic and p-hydroxybenzoic acids. The results suggest that caffeic and gallic acids may act as regulatory substances in phenylpropanoid metabolism in higher plants. Inhibition experiments with synthetic cinnamic acid derivatives have revealed that the presence of a hydrophobic aromatic ring, α,β-double bond and carboxyl group is essential for inhibitory activity. 2-Naphthoic acid which fulfills these structural requirements showed a strong inhibition. The size and shape of the active site is discussed from structure-activity relationships of cinnamic acid derivatives. o-Chlorocinnamic acid, one of the strongest inhibitors found in this study showed an inhibitory effect on the growth of the roots of rice seedlings.

      Apart from transcriptional, translational and post-translational regulations, in vitro and in vivo experiments demonstrate that many pathway intermediates or metabolites substantially affect the metabolic flux through enzyme inhibition to fine tune the monolignol biosynthetic pathway. Early studies suggested that trans-cinnamic acid and caffeic acid are the competitive inhibitors of PAL, which thereby regulate the entry of phenylalanine into phenylpropanoid and monolignol biosynthesis (Blount et al., 2000; Lamb, 1979; Sato, Kiuchi, & Sankawa, 1982).       Ocimum extracts caused epidermal necrotic area cells of cortex with the small cell size and by compaction in its cell layers, metaxylem was smaller, lateral roots formations were inhibited as compared with untreated check with water.       Sounds likely tCA positively affect gene expression of terpene biosynthesis and negatively influence the gene expression of phenylpropanoids. Although the inhibitory role of tCA on phenylpanoids pathway is well documented (Sato et al., 1982), but still more work is needed to uncover the mechanism behind the regulatory effect of tCA on the biosynthetic pathway of terpenes. Untargeted metabolomics and transcriptomics could be considered to pave the way in this issue to unravel the regulatory mechanisms of tCA and its effect on PAL activity.
