A simple enzymatic method for the preparation of radiolabeled erucoyl-CoA and other long-chain fatty acyl-CoAs and their characterization by mass spectrometry

Abstract

A simple two-step method for the biosynthesis of radiolabeled erucoyl-coenzyme A of high specific activity and other long-chain fatty acyl-coenzyme A (acyl-CoA) thioesters is reported. 1-14C-labeled erucic and oleic acids, as well as unlabeled ricinoleic and nervonic acids, were incubated at 35°C with coenzyme A in the presence of ATP, MgCl2, and acyl-CoA synthetase (EC 6.2.1.3) from Pseudomonas spp. to yield the corresponding CoA thioesters. Following incubation, each thioester was purified by rapid passage through a disposable reverse-phase C18 extraction column. The overall yields were >90% and the purities >95%, based on the distribution of radioactivity, and chromatographic and spectral properties. Fast ion bombardment-mass spectrometry was employed to confirm the structures of the various acyl-CoAs.

    Non-radioactive acyl-CoAs and sn-1-oleoyl-sn-glycerol 3-phosphate (18:1-LPA) were obtained from Sigma–Aldrich Corp. Radioactively labelled [14C]sn-1-oleoyl-sn-glycerol 3-phosphate (55 mCi/mmol) was purchased from American Radioactive Corp. 18:1-OH-CoA was synthesized following the enzymatic method by Taylor et al. [45] using the acyl-CoA synthetase from Pseudomonas sp. (Sigma–Aldrich). Analysis and quantification of the purified acyl-CoA product were performed by HPLC, using 18:1-CoA as standard.

    Peaks were identified from the retention times of acyl-CoA standards. The acyl-CoA standard for DPAn-6 was prepared from the free fatty acid and free CoA by an enzymatic method [45]. Endogenous acyl-CoA concentrations (nmol/g brain) were calculated by direct proportional comparison with the peak area of the 17:0-CoA internal standard.
