Effects of chronic insulin treatment on blood pressure in rats

Efecto de la administración crónica de insulina sobre la presión arterial en rata

Abstract

There is an evident epidemiological association between plasma insulin levels and blood pressure. The mechanism that relates insulin to blood pressure and the role of insulin in the pathogenesis of arterial hypertension have not been clearly defined. The present study was designed to examine the effects of chronic hyperinsulinism on blood pressure and to determine different related morphological variables. Wistar-Kyoto rats were subcutaneously injected with insulin (25 UI/Kg of weight) daily during the eight weeks of the experiment. Data were collected on systolic and diastolic arterial pressures and heart rate by plethysmography and direct recording (in the last week), and on morphological variables. A statistically significant elevation of systolic arterial pressure was produced after the sixth week of hyperinsulinaemia. At the end of the treatment, the systolic arterial pressure was 173.7±26.1 in the hyperinsulinaemic rats versus 153.09±21.7 in the control group. The values obtained by direct recording and by plethysmography did not differ. These results indicate that chronic hyperinsulinism produces a significant elevation in systolic blood pressure levels in the rats studied.

Resumen

Hay una clara asociación epidemiológica entre los niveles plasmáticos de insulina y la presión arterial. El mecanismo que relaciona insulina y presión arterial y el papel de la hormona en la patogénesis de la hipertensión no está bien definido. El objetivo de este trabajo consiste en examinar los efectos del hiperinsulinismo crónico sobre la presión arterial y variables morfológicas relacionadas. A ratas Wistar-Kyoto se les inyecta diariamente insulina (25 UI/Kg) por vía subcutánea durante 8 semanas. Se mide la presión arterial y la frecuencia cardíaca por pletismografía y por registro directo (última semana) y las variables morfológicas. Se observa una elevación significativa de la presión sistólica arterial tras 6 semanas de tratamiento. Al final del tratamiento, la presión sistólica arterial es de 173,7±2,1 en el grupo hiperinsulinímico versus 153, 09±21,7 en el control. Los valores obtenidos por registro directo y por pletismografía no difieren. Los resultados indican que el hiperinsulinismo crónico produce un significativo aumento en el valor de la presión sistólica arterial en la rata.
