The esterase activity between pH 5.0 and 8.5 and the protein content of splenic extracts as well as the spleen weight of 500 R whole body X-ray irradiated rats were studied from the 1–18th day after irradiation. The results were checked for statistical significance.

The esterase aotivity/mg protein is significantly increased for pH 5.0 and 6.0 from the 1–9th day and for pH 6.5–8.5 from the 2–9th day. It is significantly decreased for pH 6.0–6.5 on the 18th day. The protein concentration of splenic extracts is significantly increased on the 1st day and significantly decreased from the 6–15th day. The weight of the spleen is significantly decreased, as compared with controls, from the 1–9th day and significantly increased on the 18th day after irradiation.

As regards the morphologic changes after irradiation, the results may be applied to individual splenic cells. There are decreases in cell size and in content and concentration of cell protein. The concentration of cell protein is maintained the longest and requires the most time for restoration. The content of cell esterase is decreased for all pH values from the 1–9th day, with the exception of an increase for pH 5.0 and 6.0 on the 1st and 2nd day after irradiation. The concentration of cell esterase is increased for all pH values from the 1–3rd day after irradiation, a result of the loss of non-esterase active protein from the cells. After the 3rd day it is decreased until the 9th day. The destroyed splenic cells contain distinct protein and esterase. The surviving cells of the spleen are richer in protein and show more esterase activity in their cell protein than do the destroyed cells.

The experiments show that biochemical results of studies on single cells of the irradiated rat spleen run parallel with corresponding histological and histochemical findings after irradiation.

Es werden die Esteraseaktivität zwischen pH 5,0 und 8,5 und der Proteingehalt von Milzextrakten sowie das Milzgewicht von 500 R-röntgenganzkörperbestrahlten Ratten vom 1.–18. Tag nach Bestrahlung untersucht und die Ergebnisse auf ihre statistische Signifikanz überprüft.

Die Esteraseaktivität/mg Protein ist für pH 5,0–6,0 vom 1.–9. Tag und für pH 6,5 bis 8,5 vom 2.–9. Tag nach Bestrahlung erhöht. Für pH 6,0–6,5 ist sie am 18. Tag erniedrigt. Die Proteinkonzentration der Milzextrakte ist am 1. Tag erhöht und vom 6.–15. Tag erniedrigt. Das Milzgewicht ist vom 1.–9. Tag gegenüber dem Normalwert erniedrigt und am 18. Tag nach Bestrahlung erhöht. Alle Werte sind statistisch signifikant.

Die Ergebnisse werden unter Berücksichtigung des morphologischen Bildes nach Bestrahlung auf die Organeinzelzellen übertragen. Es treten Verminderungen der Zellgröße, des Zellproteingehalts und der Zellproteinkonzentration auf. Die Zellproteinkonzentration wird einerseits am längsten aufrechterhalten, braucht andererseits aber auch am längsten zur Wiederherstellung. Der Zellesterasegehalt ist mit Ausnahme einer Erhöhung für pH 5,0 und 6,0 am 1. und 2. Tag nach Bestrahlung für die übrigen pH-Werte vom 1.–9. Tag dauernd erniedrigt. Die Zellesterasekonzentration ist infolge stärkeren Verlustes nicht esteraseaktiver Proteine aus der Zelle für alle pH-Werte vom 1.–3. Tag nach Bestrahlung erhöht, dann aber ebenfalls bis zum 9. Tag erniedrigt. Die untergegangenen Milzzellen weisen einen deutlichen Protein- und Esterasegehalt auf. Die überlebenden Milzzellen sind proteinreicher und zeigen einen höheren Anteil an Esteraseaktivität in ihrem Zellprotein als die untergegangenen Zellen.

Die Untersuchungen zeigen, daß die Übertragung biochemischer Befunde auf die einzelnen Organzellen in der Rattenmilz nach Bestrahlung zu Ergebnissen führt, die mit den entsprechenden histologischen und histochemischen Befunden in guter zeitlicher übereinstimmung stehen.
