Summary
The efficiency of expression for foreign genes may be increased not only on the transcriptional but also posttranscriptional level. Using viral genetic systems which replicate independently on the host, it seems to be possible to increase the amount of gene product for several orders of magnitude in comparison with the hitherto experimentally reachable level.
The basic idea consists, on the one hand, in the stable transfer and expression of gene information for the produktion of functionally active RNA-dependent RNA polymerase and, on the other hand, in altering the mRNA, which encodes for the gene product in question that it exhibits the recognition sequences for the viral replicase. In this way, finally, the desired mRNA will be amplificated.
