Zusammenfassung
Drei verschiedene Diuretica —Hydrochlorothiazid, Furosemid und der diuretisch aktive Metabolit desMefrusid — wurden mit einer Mikroinfusionspumpe am Ende des proximalen Convoluts in die Henlesche Schleife einzelner Nephrone der Rattenniere infundiert. Am Anfang des distalen Convoluts wurde Tubulusflüssigkeit abgesaugt und chemisch analysiert.
Die frühdistale Natriumkonzentration steigt nach Hydrochlorothiazid schwach, nach Furosemid und dem Mefrusid-Metaboliten signifikant an. Die Konzentration von14C-markiertem Inulin bleibt nach Hydrochlorothiazid und dem Mefrusid-Metaboliten unverändert und sinkt signifikant nach Furosemid.Mefrusid und Furosemid hemmen demnach die Natriumresorption im Bereich der Henleschen Schleife. Als Wirkungsort wird der aufsteigende Schenkel angenommen. Ein entsprechender Angriffspunkt für Hydrochlorothiazid konnte weder nachgewiesen noch ausgeschlossen werden.
Summary
Three different diuretics — hydrochlorothiazide, furosemide and the active metabolite of mefruside — were infused intraluminally at the end of the proximal convoluted tubule into the loops of henle from single nephrons of the rat's kidney. At the beginning of the distal convoluted tubule tubular fluid was collected and chemicaly analyzed. The early distal sodium concentration rises only slightly after hydrochlorothiazide and increases significantly after mefruside and furosemide. The concentration of14C-labeled inuline remained unaffected by hydrochlorothiazide and mefruside and decreased after furosemide.Thus mefruside and furosemide inhibit sodium reabsorption in the loop of henle. This takes place in the ascending limb. A similar site of action for hydrochlorothiazide could be neither proven nor exluded.
