Zusammenfassung
Durch Verfütterung von Dihydrotachysterin wurde experimentell eine metastatische Verkalkung hervorgerufen.
Bei dieser Calcifizierungsform zeigten sich elektronenmikroskopisch extra- und intracelluläre Kalkablagerungen. Unter den intracellulären Veränderungen war die intramitochondriale Ablagerung von elektronendichtem Material besonders interessant: In diesen Ablagerungen konnte mittels Röntgenmikroanalyse Calcium und Phosphat direkt nachgewiesen werden.
Die Beladung der Mitochondrien mit Calciumphosphat ist eine aktive, respirationsgebundene Leistung der Mitochondrien. Sie ist mit der Funktion der Atmungskette innerhalb der „Elektronen-Transport-Partikel“ an der Oberfläche der Cristae mitochondriales verknüpft, wie sich aus Versuchen mit isolierten Mitochondrien ergab. Die in Einzelheiten beschriebene intramitochondriale Calcifizierung ist unseres Erachtens ein charakteristischer pathophysiologischer Vorgang bei der metastatischen Gewebsverkalkung, die nach unseren Befunden auch ohne primäre Zellschädigung in Gang kommen kann.
Summary
Calcification in experimental animals was started by feeding Dihydrotachysterin per os. During calcification it could be shown on an electronmicroscopic level, that calcification occurs intra- and extracellular.
In our experiments especially the intramitochondrial calcification was investigated. Using x-ray-microanalysis calcium and phosphate could directly be shown.
The uptake of calcium phosphate by mitochondria is an active process, which is directly connected with respiration and with intact electron transport particles on the surface of the cristae mitochondriales.
The intramitochondrial calcification seems to be a very typical process during reactive calcification of several tissues. This reactive type of calcification may occur without primary lesion of the cell.
