Zusammenfassung
1. Es werden Untersuchungen über den Desoxyribonucleinsäuregehalt aufgrund quantitativer feulgenphotometrischer Messungen an den Zellen der normalen Granulopoese, bei der chronischen Myelose und bei akuten Leukosen vorgelegt.
2. In den reifen Granulocyten findet sich ein ähnlicher DNS-Gehalt wie in Lymphocyten. Er liegt jedoch bei den segmentkernigen niedriger als bei den stabkernigen Granulocyten.
3. Eine Erhöhung des durchschnittlichen DNS-Gehaltes bei unreifen teilungsfähigen Granulocyten ist durch einen größeren Anteil der Zellen bedingt, die infolge prämitotischer Verdopplung des Kernchromatinmaterials einen höheren bis fast verdoppelten DNS-Gehalt aufweisen.
4. Die Befunde bei chronischer Myelose zeigen keine Unterschiede gegenüber der normalen Granulopoese.
5. Bei akuter Leukämie läßt sich bei drei von acht untersuchten Kranken in allen Paramyeloblasten eine geringe Erhöhung des DNS-Gehaltes nachweisen. Dieser Befund steht in Übereinstimmung mit Chromosomenuntersuchungen aus der Literatur, wonach sich Aneuploiden nicht nur bei Turmoren, sondern auch bei akuten Leukämien finden. Dieser Befund wird als weiterer Hinweis für die Tumornatur der Leukämiezelle aufgefaßt.
6. Im Gegensatz zu normalen Granulocytenvorstufen treten bei Paramyeloblasten nur ganz vereinzelt Zellen mit einem stark erhöhten DNS-Gehalt auf, die einen Hinweis für eine prämitotischen DNS-Verdopplung geben. Danach muß man bei den untersuchten Paramyeloblasten eine gegenüber normalen Granulocytenvorstufen verlängerte Überlebenszeit annehmen.
Summary
1. Findings are reported in connection with the DNA content of single nuclei in normal granulopoiesis in chronic myeloid and in acute leukemia after Feulgen-photometric examinations.
2. Mature granulocytes show similar DNA values as lymphocytes. The DNA values of polymorphs are somewhat lower than the DAN values of band cells.
3. The mass of normal immature granulocytes reveals diploid DNA values as mature granulocytes. But there are some immature cells which have higher or tetraploid pattern. These cells are presumed to be in interphase DNA reduplication.
4. The immature cells in chronic myeloid leukemia show nearly the same DNA pattern as normal granulopoietic cells.
5. The paramyeloblasts in 3 out of 8 patients examined are marked by a higher than diploid DNA pattern. These results are in agreement with aneuploid chromosome findings in acute leukemia by others.
6. In contrast to normal immature granulocytes in paramyeloblasts there are only single cells with higher or tetraploid DNA values, which indicate an interphase DNA reduplication. These points at a prolonged generation time of paramyeloblasts.
