Summary
In an investigation of phenolic compounds in milled grains ofZea mays, nine peaks were obtained in an HPLC chromatogram after clean-up with polyamide. The structures of these substances were established using different chemical and spectroscopic methods: Vanillic acid, caffeic acid,p-coumaric acid and ferulic acid were identified by UV spectra and retention times. After isolation by preparative HPLC, the structures of 7-hydroxy-2-oxindole-3-acetic acid and 1-O-trans-p-coumaroylglycerol were elucidated by nuclear magnetic resonance, mass spectrometry and gaschromatography (GC/MS). The oxindole derivative is known only in maize; the glycerol ester has not been detected in foods and beverages so far. Three more esters of glycerol, 2-O-p-coumaroyl-glycerol, 1-O-caffeoylglycerol and 2-O-caffeoylglycerol, were identified after semipreparative isolation by the mass spectra of their derivatives using GC/MS. The esters were synthesised and their identity manifested by co-chromatography. 2-O-Coumaroylglycerol and 2-O-caffeoylglycerol are described for the first time. Acids and glycerol esters were determined quantitatively.
Zusammenfassung
Bei der Untersuchung französischer Maisgrits auf phenolische Inhaltsstoffe wurden nach Reinigung an Polyamid im HPLC-Chromatogramm Peaks von 9 Substanzen erhalten, deren Strukturen mit Hilfe chemischer und spektroskopischer Methoden aufgeklärt werden konnten. Vanillinsäure, Kaffeesäure,p-Cumarsäure und Ferulasäure wurden über UV-Spektren und Cochromatographie identifiziert. Nach Isolierung mittels präparativer HPLC konnten mit Hilfe von NMR, MS und GC/MS die Strukturen von 7-Hydroxy-2-oxindol-3-essigsäure und 1-O-trans-p-Cumaroylglycerol aufgeklärt werden. Das Oxindolderivat ist nur im Mais bekannt, der Glycerinester wurde bisher noch nicht in Lebensmitteln nachgewiesen. Drei weitere Ester des Glycerins, nämlich 2-O-p-Cumaroylglycerol, 1-O-Caffeoylglycerol und 2-O-Caffeoylglycerol konnten nach semipräparativer Isolierung über die Massenspektren der Derivative identifiziert werden. Die Ester wurden synthetisiert und ihre Identität durch Cochromatographie gesichert. 2-O-p-Cumaroylglycerol und 2-O-Caffeoylglycerol werden erstmals beschrieben. Die Säuren und Glycerinester wurden quantitativ bestimmt.
