Summary
The influence of the NaHCO3-concentration in the culture medium on the membrane potentials of cells from different permanent strains has been investigated. In Eagle-Dulbecco Medium all cells had a membrane potential of about −52 mV. Reduction of [HCO3 −] from 44 mM to 6 mM (conditions for Hepes- and Tricin-buffer) decreased the membrane potential by about 20 mV in the case of BICR/M1R-K- and 3T3-cells. In KB- and HeLa-cells the membrane potential decreased only by about 5 mV. Replacement of Eagle-Dulbecco Medium by NaHCO3 free medium of the same pH-value resulted in a new constant membrane potential of about −33 mV within two minutes in BICR/M1R-K cells. The same decrease and the same new constant membrane potential was found, when the pH-value of the Eagle-Dulbecco Medium was adjusted to pH 6.3–6.5 by CO2 or HCOOH. The membrane properties of the investigated cells as well as the influence of different culture conditions on the membrane potential are discussed.
Zusammenfassung
Es wurde der Einfluß der NaHCO3-Konzentration des Kulturmediums auf die Höhe der Membranpotentiale von Zellen verschiedener permanenter Stämme untersucht. Alle untersuchten Zellen hatten in Eagle-Dulbecco-Medium ein Membranpotential von etwa −52 mV. Erniedrigte man in diesem Medium [HCO3 −] von 44 mM auf 6 mM (Bedingungen für Hepes- und Tricin-Puffer), so verringerte sich die Potentialdifferenz bei BICR/M1R-K- und 3T3-Zellen um etwa 20 mV. Bei KB und HeLa-Zellen betrug die Verringerung dagegen nur etwa 5 mV. Wurde das Eagle-Dulbecco-Medium gegen bicarbonatfreies Medium desselben pH-Wertes ausgetauscht, so stellte sich bei BICR/M1R-K-Zellen innerhalb von 2 min ein neues konstantes Membranpotential von etwa −33 mV ein. Den gleichen Abfall und das gleiche neue konstante Membranpotential beobachtete man, wenn der pH-Wert des Eagle-Dulbecco-Mediums durch Überleiten von CO2 oder HCOOH auf pH 6.3–6.5 eingestellt wurde. Die Membraneigenschaften der untersuchten Zellen sowie der Einfluß verschiedener Kulturbedingungen auf die Höhe der Membranpotentiale werden diskutiert.
