Zusammenfassung
Die Wirkung der 2-Desoxy-D-Glucose auf Atmung, aerobe und anaerobe Gärung und Einbau von 2-C14-Glycin in die Zellproteine von Asciteshepatom, von embryonalen und Knochenmarkzellen wurde untersucht. Die Desoxyglucose ruft in Krebszellen eine totale Hemmung der aeroben Gärung und des Glycineinbaus hervor, ohne die Atmung gegenüber den Kontrollversuchen mit Glucose zu verändern.
Die geringe oder fehlende Fähigkeit der Asciteszellen, das aus der Atmung stammende ATP für die Eiweißsynthese zu verwerten, wird als Ausdruck eines Atmungsdefektes, als funktionelle Manifestation der von Warburg beschriebenen “Schädigung der Atmung” angesehen.
Summary
The action of 2-desoxy-d-glucose on respiration, on aerobic and anaerobic fermentation, and on the uptake of 2-C14-glycine into cellular protein was studied on ascites hepatoma cells and on embryonal and bone marrow cells. In the tumor cells the desoxyglucose completely inhibited the aerobic fermentation and the uptake of glycine, without changing the respiration as compared with the control studies with glucose.
The slight or absent ability of the ascites cells to utilize, for protein synthesis, the ATP derived from the respiration, is regarded as an expression of a respiratory defect, it being a functional manifestation of the “disturbance of the respiration” as described by Warburg.
