Zusammenfassung
Die positive auxanographische Methode wurde so ausgebaut, daß sie sich zur Bestimmung des Auxotrophiegrades und des mehrfachen Nährstoffbedarfes bei polyauxotrophen Mutanen von Mikroorganismen (Hefen) eignet. Es werden ein mathematisch begründetes System von Nährstoffgemischen sowie ein Arbeitsvorgang empfohlen, die die Bestimmungen mit möglichst geringem Material-und Arbeitsaufwand mit hoher Effektivität durchzuführen erlauben. Das System umfaßt 36 Stoffe, die verschiedenartig gemischt, auf Filterpapierblättchen aufgesaugt, auf die Oberfläche von beimpften und mit einem Minimalnährboden beschickten Petrischalen gebracht werden. Die Lage und Intensität des Wachstums auf den verschiedenen Platten ermöglicht in den Diffusionsbereichen eine eindeutige Bestimmung des strikten und partiellen (±) Nährstoffbedarfes bis zu einer dreifachen Defizienz in zwei Versuchsphasen mit insgesamt sieben Petrischalen. Bei noch höherem Auxotrophiegrad wird in zwei Versuchsphasen entweder Eindeutigkeit (z.B. etwa 70% bei sechsfacher Defizienz) oder eine wesentliche Beschränkung hinsichtlich der Nährstoffbedürfnisse erreicht.
Die Verwendungsfähigkeit der Methode wurde an einigen interessanten Fällen von Mangelmutanten eines Sacc. cerevisiae-Stammes erwiesen. Bei einer Reihe durch Nitritbehandlung erhaltener Atmungsmutanten konnte ein zusätzlicher vier-bis siebenfacher Nährstoffbedarf (Leu, Ileu, Lys, His, Tyr, Val, Phe) nachgewiesen werden.
Summary
The positive auxanographic method (Beijerink; Pontecorvo; Ryan) for the identification of essential nutritive factor requirements of microorganisms was extended in such a way as to be suitable for determining the extent of auxotrophy and the various requirements of nutritive substances in polyauxotrophic mutants of microorganisms (different strains of yeast). A system of nutrient mixtures and a method of procedure are mathematically derived which enable the analyses to be carried out with the least possible expenditure of materials and effort and yet with great effectiveness.
The system comprises 36 substances mixed in different ways which are absorbed onto small disks of filter paper. These are placed on the surface of inoculated Petri-dishes poured with an agar-medium of minimal nutritive value. The observation of the site and intensity of growth in the areas of diffusion of the filter papers on the various agar plates permits the determination of the essential and partially essential (±) nutritional requirements. In all microorganisms requiring up to three nutritive substances these can be definitely determined by means of a two-step experiment employing altogether 7 Petri-dishes. In still higher degrees of polyauxotrophy a two-step experiment will yield definite results (e.g. in approx. 70% of microorganisms having sixfold nutritive requirements) or will at least help considerably in the search for the exact nutritional requirements.
The use of the method is demonstrated by means of several interesting cases of deficiency mutants of a strain of Sacch. cerevisiae. In several respiratory mutants obtained by treatment with nitrite additional four-to sevenfold nutritional requirements (Leu, Ileu, Lys, His, Tyr, Val, Phe.) could be shown.
