Zusammenfassung
Die Struktur der Erythrocytenmembran unter Einfluß verschiedener Fixierungsmittel bei variierten pH-Werten wurde anhand von Dünnschnitten elektronenoptisch untersucht.
Die Zellwand des bei neutraler Reaktion mit KMnO4 fixierten Erythrocyten ist nach Einbettung in „Plexigum“ bzw. Gelatine doppelt konturiert. Andere Fixierungsmittel zeigen derartige Strukturen nicht oder nur andeutungsweise.
Variation des pH-Wertes der Fixierungslösung verursacht bei der OsO4-Fixierung im alkalischen Bereich (ab pH 8) einen starken Kontrast der Zellwand. Mit zunehmendem Säuregrad des Fixierungsmilieus (OsO4 und KMnO4) vermindert sich die Darstellung der Membrankonturen.
Die Dopelstruktur nach KMnO4-Fixierung bleibt auch nach der Hämolyse erhalten; sie geht verloren bei Anwendung von Äther-Alkohol vor der Fixierung.
