Summary
In the mesenchyme respectively connective tissue of embryonic and adult rats 3 types of acid mucopolysacchrides were electronmicroscopically demonstrated with the rutheniumred-method. Typ 1 will be demonstrated before the first appearance of collagen structures in the early mesenchyme and shows even later no spatial relation to the collagen. These structures consist of central granula (400–600 Å) from which numerous fine filaments emanate up to 2000 Å wide in the surrounding.
In comparison type 2 shows a minor quantity of shorter filaments. It appears in major quantities in the embryonic and hyaline cartilage and is often arranged in a regular distance along the fibrills. On the surface of the thick fibrils in closely packed bundles the rutheniumred-positive material flows together in a 300 Å wide coat. The structure of type 3 is characterized by slightly waved dispersing 100–200 Å thick filaments. Until now they were only be found in the stroma of the cornea.
A comparison of the various glycosaminoglycans with respect to their occurrence during embryonic development, their specific appearance in various connection tissue types, their spatial configuration and their connection to other components of the connective tissues leads us to the assumption that type 1 represents hyaluronic acid, type 2 proteo-chondroitinsulfate and type 3 proteo-keratansulfate.
Zusammenfassung
Im Mesenchym bzw. Bindegewebe embryonaler und ausgewachsener Ratten wurden mit der Rutheniumrot-Methode die sauren Mukopolysaccharide elektronenmikroskopisch dargestellt. Dabei gelingt es, nach ihrer Morphologie 3 Typen zu unterscheiden. Typ 1 besteht aus einem 400–600 Å großen zentralen Granulum, von dem aus zahlreiche feine Fäden bis zu 2000 Å weit in die Umgebung ausstrahlen. Er läßt sich noch vor dem ersten Auftreten kollagener Strukturen im frühen Mesenchym von Embryonen nachweisen und zeigt auch später keine räumliche Beziehung zum Kollagen. Typ 2 weist dagegen eine geringere Zahl kürzerer Fäden auf. Er kommt in großen Mengen im embryonalen und hyalinen Knorpel vor und ist häufig in regelmäßigen Abständen entlang der Fibrillen angeordnet. An der Oberfläche dicker Fibrillen in dicht gepackten Bündeln fließt das Rutheniumrot-positive Material zu einem 300 Å breiten Mantel zusammen. Die Strukturen des Typs 3 bestehen aus leicht gewellt verlaufenden 100–200 Å dicken Filamenten. Sie konnten bisher nur im Stroma der Cornea nachgewiesen werden. Ein Vergleich der verschiedenen Glykosaminoglykane hinsichtlich ihres Auftretens in der Embryonalentwicklung ihres spezifischen Vorkommens in bestimmten Bindegewebsarten ihrer räumlichen Ausdehnung und ihrer Beziehungen zu anderen Bestandteilen des Bindegewebes, läßt vermuten, daß Typ 1 Hyaluronsäure, Typ 2 Proteo-Chondroitinsulfat und Typ 3 Proteo-Keratansulfat darstellt.
