Summary
Primary cultured heart muscle cells of chicken embryos fixed with glutaraldehyde show a distinct decrease in nuclear-ATPase from the circumference of the expiant to the periphery of the culture. The reaction product of ATPase was found especially in the “Pars fibrosa” of the nucleoli and also at the chromatin fibrils in the remainder of the nucleus. The decrease in ATPase-activity is accompanied by a progressive condensation of the network and therefore by a reduction in RNA-synthetic activity in the nuclei. The differential in the ATPase-activity is related to the respective energy requirements of nuclei at the sites of RNA-synthesis.
Zusammenfassung
Glutaraldehyd fixierte Primärkulturen von embryonalen Hühnerherzmyoblasten zeigen vom Rande des Mutterstücks zur Kulturperipherie hin einen deutlichen Aktivitätsabfall der Kern-ATPase. Das Reaktionsprodukt der ATPase wurde vornehmlich in der “Pars fibrosa” der Nukleolen und auch an den chromosomalen Fadenstrukturen des übrigen Kernraumes angetroffen. Der Reaktionsabfall der ATPase ist von einer Verdichtung des Strukturgefüges und somit einer Drosselung der RNS-Synthese der Zellkerne begleitet. Die unterschiedliche ATPase-Aktivität läßt sich mit dem jeweiligen Energiebedarf der Kerne an den RNS-Syntheseorten in Beziehung bringen.
