Summary

In experiments on the isolated intestine of the guinea-pig, the effects of morphine-like analgesics were compared with those of adrenaline and noradrenaline.

1. Adrenaline and noradrenaline reduce the output of acetylcholine from the intestinal wall as does morphine. The threshold concentration of adrenaline is about 10−8 w/v.

2. The inhibition by analgesics of contractions of the ileum on coaxial electrical stimulation is antagonized by sympatholytic drugs in the same way and to the same extent as that produced by noradrenaline.

3. These results may be explained in two ways: Either the analgesics act by liberating noradrenaline within the intestinal wall, or they occupy the same receptors as noradrenaline and act in the same way as this substance.

4. Washing out a paralysing dose of morphine leads to an excitation of the intestine which is potentiated by eserine. This excitation appears to be analogous to a symptom of withdrawal and is probably due to an increased output of acetylcholine.
