Abstract
Transcription factor EB (TFEB) regulates lysosomal biogenesis and autophagy, and its dysfunction contributes to the pathogenesis of many human diseases. However, the relationship between TFEB and tumorigenesis remains to be elucidated. The aim of this study is to evaluate the correlation of TFEB expression with the cancer cell proliferation, migration and invasion. TFEB gene was cloned into recombinant plasmid and transiently transfected into HeLa or MGC-803 cells. Cell proliferation, migration and invasion were assessed by MTT assay, wound healing assay and transwell assay respectively. MTT assay showed that expression of TFEB in HeLa cells enhanced cell proliferation. Wound healing assay indicated that expression of TFEB promoted the migration ability of HeLa cells. Furthermore, transwell assay suggested that TFEB expression promoted MGC-803 cell invasion through matrigel. Our study reveals a novel role of TFEB in regulating cancer cell growth.
