Abstract
The study of intracellular compartmentation of the photorespiratory metabolism involves a suitable method to separate chloroplasts, peroxisomes and mitochondria. Centrifugation on continuous or discontinuous gradients of sucrose is able to remove chlorophyll contamination and to purify mitochondria and peroxisomes (Tolbert et al. 1968, Nishimura et al. 1976, Chang, Huang 1981). However, these methods are time consuming and also cause severe damages to the organelles, because of high osmotic pressure.
