Isotretinoin and Oral Contraceptive Steroids

Abstract

One of the clinical manifestations of vitamin A deficiency is hyperkeratosis and thus it was natural for synthetic vitamin A dérivates to be made and tested in skin disorders where there is a disorder of keratinization. It is now known that at least two retinoids are of value in the treatment of skin diseases: 13-cis-retinoic acid (isotretinoin) in acne1 and etretinate (Tigason) in psoriasis.2 Both retinoids do have side-effects, however, and in particular teratogenic effects are noted in animals similar to those found in hypervitaminosis A. Hummler and Schupbach3 found eye abnormalities, cleft palate, craniofacial malformations and defects of the axial and appendicular skeleton in the offspring of rats, rabbits and mice given 2-4mg/kg of etretinate.
