In vitro culture of primary B cells specific to a short peptide

Abstract

We characterized the antibody repertoire specific to a short peptide (F21-40) of β-lactoglobulin (β-LG) by analyzing VH genes of B cell hybridomas prepared from BALB/c mice immunized with peptide-KLH conjugates. In an attempt to characterize the antibody repertoire elicited by F21-40, we established an in vitro culture system to obtain B cell clones specific to F21-40 and devised a protocol to determine the VH gene sequences.
