Abstract
In this study, detection of single nucleotide incorporation using the DNA pyrosequencing technique in a microfluidic flow-through device is demonstrated. Pyrosequencing is based on the detection of released pyrophosphate during DNA synthesis. The microfluidic platform allowed detection of single base incorporation, enabling base-by-base DNA sequencing. The miniaturization has lead to a 1000-fold reduction in reagent consumption compared to the present standard volume in pyrosequencing and will hopefully enable reading of longer DNA sequences.
