Abstract
Detection and identification of phytoplasmas (mycoplasma like organisms, MLOs) by conventional techniques such as DAPI staining followed by fluorescence microscopy, ELISA or biological indexing is limited by relatively low sensitivity and unspecificity of observed symptoms (Seemüller, 1989). Recently, rapid and sensitive methods of phytoplasma detection, based on the enzymatic amplification of pathogen DNA, were developed (Deng and Hiruki, 1990; Ahrens and Seemüller, 1992; Lee et al., 1993; Kirkpatrick et al., 1994). These methods were used successfully in our study for the diagnosis of two diseases observed in Poland.
