Zusammenfassung

Acyclovir, 9-(2-hydroxy-ethoxy-methyl)-guanin oder Acycloguanosin, zeichnet sich durch seine spezifische Wirkung auf Herpes simplex Virus (HSV) und Varizellen-Zoster-Virus (VZV) aus. Acyclovir (ACV) wird durch die HSV-spezifische Thymidinkinase über ACV-Mono- und -Diphosphat zum aktiven ACV-Triphosphat phosphoryliert, das in den virusbefallenen Zellen und nur dort akkumuliert. ACV-Triphosphat hemmt kompetitiv die HSV-spezifische DNA-Polymerase selektiv und führt als falscher Baustein in die Virus-DNA eingebaut zum Kettenabbruch. ACV ist weitgehend atoxisch.
