The Application of Nanotechnology in Constructing Scaffolds for Bone Tissue Engineering

Abstract

The bone tissue regeneration represents one of the biggest challenges in nowadays medicine, firstly due to a complex structure of human bones and wide range of diseases that can occur on that level. Bone tissue engineering and nanotechnology approach reveals a brand new, revolutionary era for treatment of bone diseases and regeneration of bone structures. Targeted drug delivery with the use of optimal dosage and reduced incidence of side effects and infections, and improved therapeutic outcome can be achieved by applying nanocarriers. The formulations based on the nanotechnology principles will enable minimally invasive therapy and better uptake of therapeutic agents from the bone cells. Among various types of nanoparticles, the most used in bone tissue engineering and bone diseases therapy are liposomes, polymeric nanoparticles, dendrimers, silica nanoparticles, carbon nanotubes, quantum dots, gold and bioceramic nanoparticles. Limitations of regenerative strategies include mismatch of cell proliferation and differentiation, insufficient mechanical strength of the scaffold and inadequate synthesis of extrinsic factors necessary for osteogenesis. Given these limitations, nanotechnology offers new solutions, attributed to advantages of nanoparticles as drug carriers which are the result of their size that is similar to the size of natural bone structures.

Scaffolds are extremely important in providing architectural support for cell growth. They are biomaterials used as a replacement and guide for tissue repair and regeneration in the form of bulk materials or with the incorporation of nanomaterials. Our aim is to provide an overview of potential application of nanotechnology in bone tissue engineering and manufacturing bio-mimetic scaffolds that can be able to replace native tissue while minimizing complications. In this article we will discuss about the latest developments in the context of scaffold construction combined with nanotechnology, as well as the potential to encourage osteogenic differentiation of such formulations.
