Definition

In 1942, Fuller Albright and colleagues reported three patients with manifestations of hypoparathyroidism (hypocalcemia, hyperphosphatemia) but no response to the administration of parathyroid extracts in terms of normalizing the abnormal calcium-phosphate homeostasis. In addition, these patients presented with short stature, a round face and obesity, brachydactyly with shortening of some of the metacarpals, and ectopic calcifications, a phenotype now described as Albright’s hereditary osteodystrophy (AHO). Albright and his colleagues concluded that the underlying mechanism explaining the phenotype in these patients consists, at least in part, of a resistance to PTH action and they coined the eponym pseudohypoparathyroidism (PHP) (Albright et al. 1942).
