Abstract

Studies of mice that either overexpress proteins or have disrupted expression of a specific gene(s) have proven invaluable to our understanding of the actions of the insulin-like growth factors (IGF-I and IGF-II) and their primary cell-surface receptor, the type 1 IGF receptor (IGF1R). They also have provided major insights into the actions of the type 2 IGF receptor (identical to the cation-independent mannose-6-phosphate receptor; IGF2R/ci-M6PR) and the growth-promoting potential of the insulin receptor (InR). Although investigation of mice with altered IGF binding protein (IGFBP) expression has yielded important information, it has not as yet revealed comparable information regarding the fundamental actions of these proteins.
