Abstract

Physiologically active proteins, glycoproteins and polypeptides are the objects of intensive research both in academic laboratories and in the applied molecular biology industry. This interest stems from a desire to understand the molecular relationship between structure and function and the potential use of many of these substances as biopharmaceuticals. From an analytical perspective, the principle milestones that need to be achieved in the analysis of protein biopharmaceuticals include 1) verifying the amino acid sequence of the protein, 2) demonstrating that miscoding has not occurred, 3) identifying and quantitating trace impurities such as other proteins, nucleic acids, antibiotics, surfactants and protease inhibitors and 4) establishing the presence and nature of blocking groups and other posttranslational modifications principle among which are the carbohydrate chains of glycoproteins.
