Abstract

The mechanism of biosynthesis of cellulose and other cell-wall polymers has been researched and reviewed for decades, and there have been many conflicting reports on the mode of cellulose synthesis in cell-free systems from higher plants. However, more recent reports (Delmer, 1977; Franz, 1978; Ray, 1980) indicate that this plethora of controversy on the mechanism of cellulose synthesis reflects the notion that the in vitro synthesis has not been demonstrated unequivocally. The purpose of this chapter is to evaluate critically the reports of in vitro cellulose synthesis, to outline some of the major problems to be overcome for such an in vitro synthesis, and to discuss some physiological considerations for cellulose synthesis in vivo.
