Abstract

Thrombin (EC 3.4.21.5) is a central, bioregulatory enzyme in thrombosis and hemostasis (1, 2). Thrombin activities are diverse and include fibrin clot formation (3) and stabilization (4), platelet and other cellular activation (5–7), and both positive and negative control of its own zymogen activation (8, 9). When compared to other serine proteinases of the coagulation and fibrinolytic systems, thrombin is unique in lacking the majority of its propiece (prothrombin fragment F 1.2) following zymogen activation (10, 11). Accordingly, the various specificities of thrombin must be accommodated solely within the enzyme moiety consisting of A and B chains.
