Abstract

Immunochemistry has a long history as a means for comparing conformations of proteins, polysaccharides and nucleic acids. Reactions of appropriate antibodies can detect changes associated with physiological modulation of protein structure (such as occurs on oxygenation and de-oxygenation of hemoglobin or allosteric modification of enzymes), evolutionary substitutions of amino acids, and the much larger changes associated with denaturation (reviewed by Crumpton, 1974). Antibodies have been important reagents for structural analysis of polysaccharides as well, being sensitive to conformation as well to the presence of different monosaccharide constituents and glycosidic linkages (Heidelberger, 1982 Kabat et al., 1981). Although nucleic acid immunochemistry has a shorter history, it has provided valuable reagents for studies of both purified nucleic acids and complex biological material such as whole nuclei or chromatin (reviewed in Stollar, 1975;1986). Antibodies distinguish among various nucleic acid structures by recognizing either specific bases or conformations.
