Pollinia Cryopreservation of Selected Orchid Species of Western Ghats, India

Abstract

Protocol for cryopreservation of nine orchid species pollinia of Western Ghats, namely, Acampe praemorsa, Calanthe masuca, Cymbidium bicolor, C. ensifolium, Dendrobium ovatum, Eulophia cullenii, Rhynchostylis retusa, Vanda wightii, and V. spathulata, is described. Pollen of C. masuca, C. bicolor, C. ensifolium, D. ovatum, E. cullenii, and R. retusa easily germinate in Brewbaker and Kwack (BK) medium supplemented with 10 gl−1 sucrose. An in vivo method is described for assessing pollen germination of A. praemorsa, V. spathulata, and V. wightii as their pollen respond poorly to BK medium. Pollinia of seven species hold more than 30% initial moisture content (m.c). Simple dehydration under laminar airflow for 3 h is recommended for reducing the m.c below 20% safe for successful cryopreservation of those seven species to get fruit set and viable seeds on pollination using the cryopreserved pollinia. Dehydration of 1–2 h is sufficient for D. ovatum and C. masuca. The protocol will be useful to establish pollinia cryobank of orchids for breeding and conservation of Nuclear Genetic Diversity.
