Technology for Molecular Cloning of Fungal Virulence Genes

Abstract

One approach to understanding the genetic and molecular basis of fungal pathogenicity is to clone the genes involved so that their physical structures, metabolic functions, and mechanisms of regulation can be analyzed both in vivo and in vitro. Before this goal can be achieved, molecular technology specific for filamentous fungi must be developed. An essential component of such technology is a transformation system based on a gene that is selectable in fungal pathogens. We have identified two genes that are attractive as selectable markers for genetically undeveloped fungi because they can be used to transform wild type cells, thereby eliminating the need for particular mutants and their complementary genes. In addition, we have shown that a fungal virulence gene can be cloned and expressed in a heterologous fungal host.
