Hepatitis C virus nonstructural protein NS3 unfolds viral G-quadruplex RNA structures
2022, Journal of Biological ChemistryCitation Excerpt :For instance, Levin and Patel reported that the HCV helicase bound to a duplex DNA substrate containing a 3′ overhang around 50 times tighter than a similar substrate with a 5′ overhang (33). In addition, Tai et al. and Gwack et al. independently demonstrated that effective duplex substrate–unwinding activity of NS3 helicase was dependent on the presence of a 3′ overhang (31, 32). Therefore, a 3′ overhang has a significant effect on the affinity and activity of NS3 on a partial duplex.

2012, Journal of Molecular BiologyCitation Excerpt :Nonetheless, HCV NS3 and NPH-II have become attractive model systems for the mechanistic analysis of RNA helicase activity.8 Both HCV NS3 and NPH-II unwind RNA and DNA duplexes in a unidirectional, stepwise, and processive fashion.6,8–12 This unwinding mode resembles canonical DNA helicases.13
Development of robust in vitro serine protease assay based on recombinant Pakistani HCV NS3-4A protease
2011, Virus ResearchCitation Excerpt :That is, the NS4A cofactor enhances the cleavage at NS5A/B site and is absolutely required for cleavage at the NS4B/5A site (Landro et al., 1997; Barbato et al., 1999). Besides containing a serine protease domain (one-third of NS3) of about 20 kDa at its N-terminus, it harbors RNA helicase, approximately 400 C-terminal residues (Gwack et al., 1996). Deletion experiments have shown that helicase and protease domains can work independent of each other as separate polypeptide chains expressing their respective activities (Tanji et al., 1994; Kolykhalov et al., 1994; Failla et al., 1995; Hahm et al., 1995; Kim et al., 1996).
