As shown in Fig. 3, the GlcNAc-attached IAG with insulin-type disulfides (1) showed the spectral pattern typical for insulin-family peptides [31–33]. The non-glycosylated IAG with insulin-type disulfides (2) showed almost the same spectral pattern, strongly suggest that GlcNAc moiety attached at AsnB30 did not affect the IAG peptide conformation.

We report here the first crystal structure of invertebrate insulins expressed from cloned cDNAs, namely two variants of DILP5, DB and C4, that differ by the absence or presence of an Asp-Phe-Arg sequence extension at the N terminus of the A-chain.

These proteins contain ThrB5 and ArgB5, respectively.

This 3D model of Cq-IAG includes the entire classical Cys-containing region in both chains (encompassing residues 24–54 of the B chain and 142–165 of the A chain).
