In fact, most antibiotics are ineffective against intracellular pathogens due to their low retention inside the cells (i.e. macrolides or fluoroquinolones) [7,8] or poor intracellular accumulation (i.e. β-lactams or aminoglycosides) [9,10]. In particular, vancomycin is the last resort against MRSA but cannot eradicate intracellular MRSA due to its low uptake by infected host cells [11].

The cellular gentamicin content after incubation with pifithrin-α was assessed by a disc-plate microbiological technique using Bacillus subtilis (ATCC 6633) as previously described (Tulkens and Trouet, 1978), and no statistically significant change in gentamicin accumulation was observed (data not shown). To confirm the involvement of p53 protein in gentamicin-induced apoptosis, we also performed p53 siRNA-mediated gene silencing experiments.

However, pinocytosis of gentamycin is not restricted to renal cells but up-take of gentamycin was reported to be faster and more efficient in renal cells than other cell types (Morin et al., 1980; Tulkens and Trouet, 1978). The higher uptake rate of gentamycin by renal cells is due to higher levels of the protein megalin, which binds polybasic compounds, and acidic PLs compared to other cell types (Carryn et al., 2003).
