Abstract:
Dormant axillary buds of various magnolia hybrids, 'Elizabeth', 'Yellow Bird', 'cultivar #149' and Magnolia stellata were cultured on Murashige-Skoog medium (MS), Anderson-Rhododendron medium (AR) and a medium containing lower concentrations of macronutrients than the other two.
All media were supplemented with m-inositol, 500 mg/L; Jacquoit vitamins; benzyladenine, 5–15 mg/L; and napthaleneacetic acid, 1–5 mg/L. Cultures were maintained on a 16 hour photoperiod or total darkness at 21 C for 4 weeks.
Cultures with 240 mg/L KNO3 concentration and carbohydrates of 1% at low light intensity had a greater survival rate and larger calli development.
All hybrids did better on the medium containing low concentrations of macronutrients.
