Effect of Panax Notoginseng Saponins on Extracellular Matrix Damage and Oxidative Stress After Cerebral Ischemia-reperfusion in MiceChinese Full Text

Abstract: Objective:To investigate the effect of Panax Notoginseng Saponins(PNS) on extracellular matrix damage and oxidative stress after cerebral ischemia/reperfusion injury in mice.Methods:C57BL/6 mice were randomly divided into sham-operation group,model group,PNS group and Edaravone group.The bilateral common carotid artery of mice were ligated for 20 min followed by 24 hours reperfusion after administration for 4 days,the expression of MMP-9 and TIMP-1 protein in brain tissue was detected,the content of malonaldehyde(MDA),glutathione(GSH),nitric oxide(NO) and activity of superoxide dismutase(SOD) and nitric oxide synthase(NOS) in brain tissue after cerebral ischemia for 20 min follwed reperfusion for 60 min was determined.Results:Compared to sham operated group,the expression of MMP-9 protein was in brain tissue in model group incressed markedly(P < 0.01),the expression of TIMP-1 protein had no changes.after cerebral ischemia-reperfusion,The content of MDA,NO and activity of NOS in model group was higher than the sham operated group(P < 0.01),The content of GSH and activity of SOD in model group was signifi-cantly lower than the sham operated group(P < 0.01).The expression of MMP-9 protein,the content of MDA and NO in PNS group was lower than the model group(P < 0.05,P < 0.01),the expression of TIMP-1 protein in PNS group was higher than the model group(P < 0.01),the content of GSH and activity of SOD and NOS in PNS group had no changes(P > 0.05).Conclusion:Extracellular matrix damage and oxidative stress injury after cerebral ischemia-reperfu-sion in mice could be inhibted by PNS.
