Simultaneous content analysis of flavonoids in Cirsium japonicum var. mackii using HPLC analysis

Flavonoids in Cirsium japonicum have been reported to possess many pharamacological activities e.g. anti-inflammatory, anti-cancer, anti-mutagenic, anti-fungal, enhance immunity [1]. Flavonoids such as pectolinarin, hispidulin-7-neohesperidoside, 5,7-dihydroxy-6,4'-dimethoxyflavone, linarin, and luteolin were already quantified in C. japonicum [2]. These flavonoids are the main components of C. japonicum. We isolated and identified four flavonoids, linaroside (1), apigenin (2), hispidulin (3), and pectolinarigenin (4) from C. japonicum var. mackii (Figure 1). A simultaneous analysis of four compounds in each part (leaf collected during spring (CJL1) and fall (CJL2,3), flower (CJF, CJF2), root (CJR3) dried by hot blast (CJR1) and freeze (CJR2), seed (CJS), pappus (CJP), and the whole plant (CJL4) of C. japonicum var. mackii was done. HPLC/UV analysis was conducted with a gradient solvent system of water (containing 0.5% acetic acid) and acetonitrile (ACN). ACN was decreased from 70% to 0% for 40 min by gradient elution program. UV detector was set at an absorbance of 287nm. From the result, highest concentration of linaroside (1) is contained in CJF2 (11.27 mg/g), apigenin (2) is CJF (0.46 mg/g), hispidulin (3) is CJL4 (1.95 mg/g), and CJF2 for pectolinarigenin (4) (21.51 mg/g) of C. japonicum var. mackii (Table 1). The content of flavonoids were higher in flower and leaf than in root. The identification of the abundance of flavonoids in leaf and flower compared to other parts will provide a good material for further use. The results of this experiment could be used as a standard data of each part of the content in C. japonicum var. mackii.
