Regulation of heat shock protein-70 gene transcripts in breast cancer cells during hypo and hyperthermia exposure
2019, Meta GeneCitation Excerpt :These proteins also enhance cell survival following a multitude of stresses, including elevated temperature, hypoxia, oxidative stress, altered pH, and heavy metals, etc. HSP-70 is frequently over expressed in cancer relative to normal cells, possibly due to high levels of proteotoxic stress in tumors and subsequent activation of its master regulator, heat shock factor 1 (HSF1) (Ricaniadis et al., 2001; Ramp et al., 2007; Akerfelt et al., 2010). Further, knock out studies have demonstrated its protective role against genomic instability (Hunt et al., 2004).
Recent advancement of piperidine moiety in treatment of cancer- A review
2018, European Journal of Medicinal ChemistryCitation Excerpt :HSP70 is also involved in inhibition of apoptosis and is highly expressed in malignant melanoma. However, it is under-expressed in renal cancer cells [152–154]. Zeng et al. [155] targeted heat shock protein 70 (HSP70) which has an important role in regulation of apoptosis.
Turning tumour cells into antigen presenting cells: The next step to improve cancer immunotherapy?
2016, European Journal of CancerCitation Excerpt :In these tumours, some studies found that HLA-DR expression correlated with the presence of TILs (CD4 and CD8 T cells) [37–39] and was associated with a good prognosis [40,41]. Other studies failed to demonstrate a survival advantage for HLA-DR expressing melanoma cells [42,43]. These discrepancies may be due to the fact that MHC-II expression does not always send an ‘immunogenic’ signal as suggested by the higher expression of MHC-II in metastatic lesions than in primitive lesions [37].
Regulation of molecular chaperones through post-translational modifications: Decrypting the chaperone code
2013, Biochimica et Biophysica Acta - Gene Regulatory MechanismsCitation Excerpt :Since both cochaperones are TPR-containing proteins, it is believed that they compete for chaperone binding through the EEVD motif, thereby generating an equilibrium between folding and degradation of Hsp cargo. Expression levels of Hsp70 have been shown to be either up- or down-regulated in a variety of cancer types [18,19] and although some have questioned its validity as a prognostic marker [20] it would appear that depletion or inactivation of this chaperone sensitizes tumor cells to apoptosis [21–23]. Hsp70 can also provide a shielding effect against neurodegenerative aggregation disorders such as Huntington's and Parkinson's disease, either by directly promoting resolubilization of aggregates or by preventing apoptosis of the neurons [24,25].
