Isolation of Infectious Poliovirus Ribonucleic Acid from Supernatant Fluids after Ultracentrifugation

Abstract

Huppert and Sanders1 reported earlier that after pelleting the intact encephalomyocarditis (EMC) virus in the ultracentrifuge and extracting both the pellet and supernatant with cold phenol, infectious ribonucleic acid (RNA) was obtained only from the supernatant. Recently, Colter et al.2 contradicted the above results; they found the yield of infectious RNA from EMC virus and mengo-encephalitis virus was a function of the infectivity titre of the intact virus pellet, and after removal of virus by centrifugation, infectious RNA could not be extracted from the supernatant. Herein we report results obtained with poliovirus after ultracentrifugation.
