ERK-1/2 has been found in the astrocytes of SOD1G93A animals at a late stage of disease development, showing that ERK-1/2 amplification in reactive astrocytes could be caused by motor neuron dysfunction (Chung et al., 2005; Lu et al., 2011). P2X purinoceptor 7 (P2 × 7) receptor-induced oxidative increases in ALS microglia are considered to be triggered by ERK-1/2 (Apolloni et al., 2013).

Transcriptional upregulation of Mkp1 and Mkp3 by sodium butyrate in R6/2 HD mouse models provides a putative mechanism that explains how HDAC inhibitors counteract the unopposed activation of Erk that is known to lead to ferroptotic death in vitro (Yagoda et al., 2007; Zille et al., 2017) and that has also been implicated in neurodegeneration in vivo (Chung et al., 2005; Feld et al., 2014; Pei et al., 2002; Santini et al., 2010). Moreover, sodium butyrate treatment also decreased tau expression, although this appeared to be limited to the neurons in the striatum (Ferrante et al., 2003).

Elevated levels of the inflammasome component IL-1β and phosphorylated ERK is also observed in spinal cord astrocytes of ALS mice and patients (Chung et al., 2005; Johann et al., 2015). The secretion of IL-6 by ALS astrocytes can be elicited by the pathogenic pro-inflammatory effector TWEAK (Bowerman et al., 2015), or by the cerebrospinal fluid (CSF) obtained from ALS patients, which in addition to promote TNFα production, leads to decreased astrocytic expression of GDNF by astrocytes (Mishra et al., 2016).

2016, Neuroscience and Biobehavioral ReviewsCitation Excerpt :In the hippocampus, of pre-symptomatic SOD1G93A mice we observed, an enhanced reactivity to spatial novelty associated with a higher expression of the hippocampal α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) GluR1 subunit (mRNA and protein), and with an increased induction and maintenance of CA3-CA1 LTP (Spalloni et al., 2006). These findings are further supported by molecular evidence showing increased levels of phosphorylated Erk, an extracellular signal-regulated kinase involved in neural plasticity and memory in symptomatic SOD1G93A mice (Chung et al., 2005). In a paper just published Quarta et al. (2015) described hippocampal alterations in the SOD1G93A mouse, prior to the onset of motor symptoms, affecting GABAergic interneurons (parvalbumine-positive).

2014, Journal of NeuroimmunologyCitation Excerpt :The knockout experiments described in the study reported by Tam et al. using mouse embryonic fibroblasts demonstrated that all three components, IRE1, PERK and IκB kinase, are required for the complete activation of NF-κB during ER stress (Tam et al., 2012). Western blot and immunohistochemical analyses demonstrated that the expression of p-PERK and p-eIF2α was increased in the spinal cord of transgenic SOD1G93A mice (Chung et al., 2005; Nagata et al., 2007) and SOD1G93A-expressing motor neurons (Prell et al., 2012). The mice, which carried an ALS mutation and a loss of function mutation of PERK [G85R/PERK(+/−)], exhibited a dramatically accelerated disease onset, as well as shortened disease duration and lifespan.
