GroEL/ES inhibitors containing benzothiazole and hydroxyl exhibited potent anti-bacterials against planktonic and biofilm forms of S. aureus, and leads with low cytotoxicity could permeate through established S. aureus biofilms and maintain their bactericidal effects [11–13]. While another structural motif diarylurea was an important pharmacophore for anti-bacterials in drug discovery, for example, triclocarban, PK150 and PQ401 showed sub-μM activity [14–20].

For each series, 15 left-side groups were assessed that contained a diverse range of substructures, including some that we have found effective with other GroEL/ES inhibitor scaffolds that have shown antibacterial properties (e.g. thiophenes, 2-chlorothiophenes, and aryl-sulfonamides).56–58,71 These analogs were tested in a panel of in vitro assays for bacterial proliferation inhibition, GroEL/ES inhibition (both substrate folding and ATPase functions), human cell cytotoxicity, and bacterial resistance towards lead analogs.
