A new oleanane-type triterpene from Ardisia lindleyana D.Dietr and its cytotoxic activity

A new oleanane-type triterpene, ardisiapunine A (1), together with eight known compounds were isolated from the roots of Ardisia lindleyana D.Dietr. Their chemical structures were determined by means of spectroscopic methods including HR-ESI-MS and (1 D, 2 D) NMR data. The absolute configuration of compound 1 was established by a single-crystal X-ray diffraction experiment. The new compound is an unusual oleanane-type triterpene bearing an acetal and a C-13—C-18 double bond. The cytotoxicity of all isolated compounds were evaluated using four human cancer cell lines, including A549, HepG2, HeLa and U87. The new compound 1 and compound 2 were weakly active but the known compound 6 exhibited a high cytotoxicity compared to cisplatin.
Cytotoxic alkyl benzoquinones and alkyl phenols from Ardisia virens

Bioassay-guided fractionation of roots and stems of Ardisia virens Kurz (Myrsinaceae) led to isolation of fourteen compounds, (2′R)-6-(2′-acetoxypentadecyl)-5-hydroxy-2-methoxy-1,4-benzoquinone (1), (2′R)-6-(2′-acetoxytridecyl)-5-hydroxy-2-methoxy-1,4-benzoquinone (2), (2′R)-6-(2′-acetoxytridecyl)-2-methoxy-1,4-dihydroxybenzene (3), (2′R)-6-(2′-acetoxytridecyl)-5-formyl-2-methoxy-1,4-dihydroxybenzene (4), 1-(3,5-dihydroxyphenyl)nonan-1-one (5), 1-(3-hydroxy-5-methoxyphenyl)pentan-1-one (6), (2′S)-3-hydroxy-5-methoxyphenyl-2′-tridecanol (7), (2′S)-3-hydroxy-5-methoxyphenyl-2′-pentadecanol (8), (2′S)-5-acetoxy-3-hydroxyphenyl-2′-tetradecanol (9), 1-acetoxy-2-methoxy-6-pentadecyl-4-hydroxybenzene (10), 1-acetoxy-2-methoxy-6-tridecyl-4-hydroxybenzene (11), (2′R)-1-acetoxy-6-(2′-acetoxy-tridecyl)-2-methoxy-4-hydroxybenzene (12), (2′R)-1-acetoxy-6-(2′-acetoxypentadecyl)-2-methoxy-4-hydroxybenzene (13), and 1-acetoxy-6-(2′-ketopentadecyl)-2-methoxy-4-hydroxylbenzene (14), together with thirty-four other known compounds, of which three, 1-(3,5-dihydroxyphenyl)pentan-1-one (15), 1-(3,5-dihydroxyphenyl)heptan-1-one (16), and 1-(3,5-dihydroxyphenyl)pentadecan-1-one (17), were isolated for the first time from a natural source. Their structures were elucidated by analyses of spectroscopic data. Seven cytotoxic constituents were found to be 3, 10–12, ardisianone (18), cornudentanone (19), and ardisianol (31) with cytotoxic properties (IC₅₀ values ⩽4 μg/mL) against MCF-7, NCI-H460 and SF-268 cancer cell lines in vitro.
The genus Ardisia: A novel source of health-promoting compounds and phytopharmaceuticals

Approximately 500 species of Ardisia (Myrsinaceae) are found throughout tropical and subtropical regions of the world. Several of those Ardisia species have been used as ornamental plants, food and medicines. Due to taxonomic confusions, correct identification and acquisition of plant materials remain difficult for some species. Although species of Ardisia are a rich source of novel and biologically potent phytochemical compounds, such as bergenin and ardisin, the utilization of Ardisia species or their phytochemical constituents have not been fully explored, resulting in underexploitation of their uses. The present article reviews the usage and biological activities of Ardisia compounds, as well as recent progress regarding the use of this genus in clinical research. The information presented here also illustrates the potential of the genus as a source of therapeutic agents.
Sulfidopeptide Leukotriene Receptor Binding Assays

The chapter presents a study on sulfidopeptide leukotriene receptor binding assays. The sulfidopeptide leukotriene receptor binding assay has been used extensively in antagonist compound screening, in the identification of antagonists from fermentation broths and natural products, and also for the quantification of sulfidopeptide leukotrienes in biological samples. Sulfidopeptide leukotrienes are highly lipophilic and tend to interact nonspecifically with plastics, paper, test tubes, glass, and other nonreceptor sites. The radiolabeled agonists are metabolically unstable and sensitive to chemical degradation. These are the major factors to be carefully considered when setting up radioligand binding systems to study the receptors and the mechanisms of receptor regulation. Recent advancement of the synthesis of potent receptor antagonists has alleviated some of the vexing difficulties. When appropriately controlled and validated, the radioligand binding assay has proved to be extremely useful. The major objectives of the saturation binding experiments are as follows: (1) demonstrate that the number of the radioligand binding sites is limited and thus saturable, and (2) determine the density of the receptor and the affinity of the receptor for radioligand. It is found that [³H]LTD₄ binds to the guinea pig lung-specific sites in a high affinity and saturable manner—the very first and the most important criterion of establishing the membrane receptor. Radiolabeled [³H]LTE₄ can also be used to label the sulfidopeptide leukotriene receptor sites. [³H]LTE₄ is more hydrophobic than [³H]LTD₄.
