Regulation and development of muscarinic acetylcholine receptors

Abstract

Knowledge of the regulation, development, structure, and function of neurotransmitter receptors is crucial in order to have a detailed understanding of synaptic transmission. The nicotinic acetylcholine receptor has served as an invaluable model for the study, characterization, purification, and reconstitution of a neurotransmitter receptor. Biochemical and molecular investigations have now begun to elucidate the action and regulation of muscarinic acetylcholine receptors as well.

      In particular M1, M3 and M5 subtypes, activate phosphoinositide hydrolysis, while M2 and M4 subtypes inhibit adenylyl cyclase, leading to a decrease of cAMP level. Moreover, they can directly modulate potassium, calcium and chloride channels [19,23,25]. Muscarinic modulation of second messengers such as cGMP and cAMP has already been reported in DRG neurons [6,29].
