While some evidence of processing of pre-tRNA by exoribonucleolytic cleavage for their 3′-end formation was originally obtained in bacteria in the 1970s [224], subsequently it was concluded that mature tRNAs were formed by downstream cleavage followed by 3′ end formation by precise exoribonucleolytic trimming [13]. Independent evidence for the maturation of tRNA 3′-ends by a single endoribonucleolytic cleavage reaction came from eukaryotes from assays of tRNA in extracts prepared from germinal vesicles of Xenopus laevis [225,226].

RNase P cuts within intergenic regions of several polycistronic RNAs including tnaAB, rbsDACBK, lacZYA, secG‐leuU, and hisG‐I (203–205).
