Infectious viral RNA was prepared by treating a virion suspension with proteinase K and SDS at 60°C for 2 min, and viral RNA was recovered by phenol–chloroform extractions as described by Carrington and Morris (1984). Virus- and mock-inoculated tobacco leaves were rinsed with tap water, immersed in 70% ethanol for 30 s, and rinsed again with water.
