2023, International Journal of PharmaceuticsEvaluation of release and pharmacokinetics of hexadecylphosphocholine (miltefosine) in phosphatidyldiglycerol-based thermosensitive liposomes
2021, Biochimica et Biophysica Acta - BiomembranesCitation Excerpt :Thermosensitive liposomes (TSL) belong to a class of stimuli-responsive nanocarriers which can release encapsulated compounds at temperatures in the mild hyperthermia (HT) range (41°-43 °C) [17,18]. Although the release mechanism might slightly differ between TSL formulations, it is generally assumed that drug release predominantly occurs at boundary regions between solid-gel and liquid-crystalline phases which are formed in the TSL bilayer upon heating to Tm [19–21]. This provides a possibility for the intravascular, HT-induced drug release from TSL resulting in improved tumor accumulation of the bioavailable drug [22,23].
Design, evaluation and in vivo pharmacokinetic study of a cationic flexible liposomes for enhanced transdermal delivery of pramipexole
2021, Journal of Drug Delivery Science and TechnologyCitation Excerpt :It is believed that the molecular mobility of phospholipid increases gradually at higher temperatures approaching phase transition temperature. This leads to improved lipid bilayer permeability [53], resulting in rapid drug leakage. Other contributing factors of drug leakage are the higher partition coefficient of pramipexole [54,55] and the concentration gradient across the lipid bilayer.
Hyperthermia can alter tumor physiology and improve chemo- and radio-therapy efficacy
2020, Advanced Drug Delivery ReviewsCitation Excerpt :Targeted, heat-triggered release is of particular interest in oncology where toxic ChT agents can have debilitating off-target effects in normal tissue. TSL development has been built on decades of research characterizing the temperature-dependent permeability of lipid bilayers [351,352] and liposomal drug carriers [353,354]. Lipid bilayers have a characteristic temperature known as the melting temperature (Tm), above which the lipids exhibit fluidity of movement and below which the bilayer is referred to as solid with comparatively little displacement of the lipids.
