2023, Industrial Application of Functional Foods, Ingredients and Nutraceuticals: Extraction, Processing and Formulation of Bioactive CompoundsDesign, synthesis, biological evaluation and molecular modelling studies of indole glyoxylamides as a new class of potential pancreatic lipase inhibitors
2019, Bioorganic ChemistryCitation Excerpt :Amongst the several targets that have been explored to treat or prevent obesity [6–13], pancreatic lipase (PL) is considered as a successful target due to its tolerable side effects. Human PL (EC 3.1.1.3), encoded by the PNLIP gene, is a primary digestive enzyme secreted from the pancreatic exocrine [14,15], and is primarily involved in the hydrolysis of triglycerides [16]. The crystal structure of human PL (PDB ID: 1LPB) is composed of 449 amino acids, with the catalytic triad Ser 152 - Asp 176 - His 263, defining the active site of the enzyme [17].
Changes of the expressions of multiple metabolism genes in rat pancreatic islets after ventromedial hypothalamic lesioning
2015, Neuroscience LettersCitation Excerpt :In the present study, real-time PCR analysis also demonstrated that VMH lesions downregulated the Pnlip gene. Pnlip is a protein secreted by the acinar pancreas and is essential for the hydrolysis and absorption of long-chain triglyceride fatty acids in the intestine [1]. Consistent with this, in the pancreatic islets of AKR/J rats and C57BL/6J mice, the elevated RNA expression level of exocrine pancreatic proteins was detected [4].
Identification of a novel mutation in the PNLIP gene in two brothers with congenital pancreatic lipase deficiency
2014, Journal of Lipid ResearchCitation Excerpt :Genomic DNA was extracted from peripheral leukocytes following standard protocols. DNA was amplified to obtain all PNLIP (4) and CLPS (5) genes coding exons and their flanking regions using conventional PCR techniques. PCR products were purified using magnetic particle technology (Seradyn, Inc.).
